A new preprint by Stavros Therianos leverages the first complete synaptic wiring diagram of an insect brain—the larval Drosophila connectome—to address a fundamental question in neuroscience: how much of a neural circuit's behavior is fixed by its wiring diagram versus the unrecorded physiological details at each synapse? The authors ran this complete connectome as a fixed, rate-based dynamical model where no single-neuron parameters were tuned, so any emergent dynamics arise purely from the wiring and its connection strengths. They then compared the model's behavior against a hierarchy of randomized networks that preserved coarser descriptions of the wiring, from degree distribution to overall synapse counts.
The results reveal a clear separation: the overall dynamical regime—how strongly and richly the network responds—is driven by statistical wiring properties such as the distribution of connections. Randomized networks that preserved only these coarse statistics replicated the model's global responsiveness. However, the precise pattern of connections beyond these statistics is what determines the geometric trajectory of activity. For example, sparse input is tightly confined to a compact olfactory pathway in the real connectome, but it floods broadly across randomized networks. Furthermore, the mushroom body (the insect learning center) plays an outsized role in shaping recurrent dynamics, appearing prominently in leading adjoint-side modes. This separation clarifies which connectome-based claims are supported by wiring alone, offering a powerful framework for interpreting future connectome studies in larger brains.
